Retatrutide is a single peptide that consists of 39 amino acids engineered from a GIP peptide backbone to stimulate GLP-1, GIP, and GCGRs (Fig. 1). There are three non-coded amino acid residues [two α-amino isobutyric acids (Aib), and one α-methyl-L-leucine (αMeL)] at positions 2, 20, and 13 of the peptide backbone.
